Título: Influência da melatonina exógena sobre parâmetros sanguíneos, hormonais, metabólicos e inflamatórios em ratas alimentadas com dieta hipercalórica durante a prenhez

Resumo: Atualmente tem-se buscado estratégias terapêuticas como a melatonina para abrandar os sintomas das comorbidades relacionadas à obesidade, pois esta tem sido associada à redução do peso corporal. Assim, a presente pesquisa objetivou avaliar parâmetros metabólicos, bioquímicos e histoquímicos no fígado, bem como analisar a interação da melatonina com a histofisiologia renal e o sistema imune em ratas prenhes alimentadas com dieta hipercalórica associada ou não a esse hormônio. Utilizou-se 40 ratas prenhes divididas nos grupos (n=10): I: ratas submetidas à dieta comum (D); II: ratas submetidas à dieta hipercalórica (DH); III: ratas submetidas à dieta hipercalórica + melatonina (0,5 mg/kg) (DH+Mel) e lV: ratas submetidas à dieta comum + melatonina (0,5 mg/kg) (D+Mel). Os resultados revelaram que ao 7° dia as fêmeas dos grupos DH e DH+Mel apresentaram ganho de peso corporal. Ao 14° e 20° dia, apenas as fêmeas do grupo DH mantiveram essa condição. Não houve diferenças para a ingestão sólida e líquida, ao 7° dia. Ao 14° e 20° dia, houve elevação desse parâmetro nas fêmeas do grupo DH. Os níveis séricos de ALT, AST, GAMA GT, fosfatase alcalina, amilase, lipase, frutosamina, colesterol total, LDL, triglicérides, HDL, glicose e insulina aumentaram aos 7° e 14°dia nas fêmeas do grupo DH, com exceção dos níveis de amilase que não diferiu entre as fêmeas dos grupos (7° dia) e redução da FA e HDL (14°dia). Nas fêmeas dos grupos tratados com melatonina os níveis de amilase foram reduzidos. Ao 20° dia, as fêmeas do grupo DH apresentaram o mesmo comportamento observado ao 14° dia. Porém as fêmeas que receberam a melatonina exibiram aumento da FA e do HDL, enquanto os demais parâmetros apresentaram valores reduzidos em relação aos grupos D e DH. A quantidade de lipídio no fígado, bem como o teor de colágeno foram significativamente elevados nas fêmeas do grupo DH em relação aos demais grupos. Nas fêmeas do grupo DH+Mel houve diminuição significativa na quantidade de gordura e de colágeno. Nas fêmeas dos grupos D, DH+Mel e D+Mel não ocorreram diferenças em relação à quantidade de glicogênio hepático, diferindo das fêmeas do grupo DH que apresentou níveis elevados em relação a estes. Observou-se que as fêmeas do grupo DH apresentaram alterações hematológicas, urinárias e bioquímicas significativas, melhorando os níveis de uréia, creatinina, glicose, bilirrubina, leucócitos, e plaquetas, sendo tais níveis normalizados pela administração da melatonina. Além disso, a marcação de colágeno e citocinas proinflamatórias no baço (IL-6 e TNF-α) foi elevada e a de VEGF reduzida. A melatonina exógena mostrou ser uma alternativa terapêutica eficaz, apresentando vários efeitos antiobesogênicos durante a prenhez em ratas e reduzindo os efeitos adversos causados pela obesidade.
Abstract: Nowadays researchers have tried treatment strategies such as melatonin to slow down the symptoms of comorbidities related to obesity, since it has been associated with reduced body weight. Thus, the present study aimed to evaluate metabolic, biochemical and immunohistochemical parameters in the liver, as well as analyzing the interaction of melatonin with renal histophysiology and the immune system in pregnant rats fed with high calorie diet with or without this hormone.We used 40 pregnant rats divided in the groups (n=10): I: rats subjected to the common diet (D); II: rats submitted to high calorie diet (DH); III: rats submitted to high calorie diet + melatonin (0.5 mg / kg) (DH + Mel) and IV: rats subjected to the common diet + melatonin (0.5 mg / kg) (D + Mel). The results showed that at day 7 females of DH and DH + Mel groups showed body weight gain. At 14th and 20st day, only the females from DH group retained this condition. There were no differences for the solid and liquid intake, in the 7th day. At 14th and 20st day, there was an increase of this parameter in females of DH group. The serum levels of ALT, AST, Gamma GT, alkaline phosphatase, amylase, lipase, fructosamine, total cholesterol, LDL, triglycerides, HDL, glucose and insulin increased at 7th and 14th day in females of DH group, with the exception of amylase levels that did not differ between females of the groups (day 7) and reduced HDL and AF (day 14). In females of the groups treated with melatonin, amylase levels were reduced. At 20st day, the females of DH group showed the same behavior observed at 14 days. However, females who received melatonin showed increased FA and HDL, while the other parameters showed reduced values on the D and DH groups. The amount of lipids in the liver as well as the collagen content was significantly higher in females of DH group compared to other groups. In females of DH + Mel group there was a significant decrease in the amount of fat and collagen. In females of groups D, DH + Meland D + Mel there were no differences in relation to the amount of hepatic glycogen, differing from the females of DH group that had elevated levels relative to these.It was observed that females from DH group had hematological, urine and biochemical significant changes, as the improve levels of urea, creatinine, glucose, bilirubin, leukocytes, and platelets, and these levels normalized by administration of melatonin. Furthermore, the marking of collagen and proinflammatory cytokines in spleen (IL-6 and TNF-α) was elevated and reduced VEGF. Exogenous melatonin was shown to be an effective alternative therapy, with various anti-obesity effects in rats during pregnancy and reducing the adverse effects caused by obesity.
